离子络合剂及常见阳离子对博莱霉素作用淋巴细胞SCE的影响
Effects of some cations and their chelating agents on lymphocyte SCE treated by bleomycin
【摘要】 目的观察几种离子和离子络合剂对博莱霉素作用淋巴细胞SCE的影响。方法在含有一定量BLM的人外周血淋巴细胞培养物中注入一定量的几种金属离子或离子络合剂,统计淋巴细胞的姐妹染色单体交换率(sister chromatid exchanges,SCE率)的变化,以观察这些离子对BLM的影响作用。结果终浓度为0.5和1.0mmol/L的Fe2+、终浓度为2.0和4.0 mmol/L的Mg2+在BLM切割DNA上有明显的促进作用(P<0.05);终浓度为2.0 mmol/L的Fe2+则有极其明显的促进作用(P<0.01);同为2.5 mmol/L的EDTA和枸橼酸钠(SC)则有明显的拮抗作用(P<0.05)。结论一定量的Fe2+和Mg2+的存在对于BLM切割DNA的过程是必需的,EDTA和枸橼酸钠的络合作用则从反面印证了该结论。
【Abstract】 Objective: To observe the effects of some cations and their chelating agents on lymphocyte SCE treated by bleomycin.Methods: SCE(sister chromatid exchanges) were observed and contrasted when catholytes,chelating agents or bleomycin had been added into the culture of human blood lymphocytes.Results: The groups of ferrous chloride(0.5 or 1.0mmol/L) and magnesium chloride(2.0 or 4.0mmol/L) show a significant difference from the control(P<0.05);The group of ferrous chloride(2.0mmol/L) show a very significant difference from the control(P<0.01);Sodium Citrate and EDTA(2.5mmol/L both) show a significant difference from the control(P<0.05).Conclusions: Ferrous ion and magnesium ion are both necessary to the action of bleomycin on lymcytes SCE.
