NKCC1基因敲除小鼠的饲养繁殖及其鉴定
Breeding,Reproducing,and Identifying NKCC1 Knock-out Mice
【摘要】 目的繁殖和鉴定NKCC1基因敲除小鼠,建立实验动物模型。方法将引进的NKCC1基因敲除小鼠饲养于相对无噪声的隔离环境中,依照遗传学规则进行繁殖及培育,提取小鼠尾部组织全基因组DNA进行聚合酶链式反应(PCR)检测各种系小鼠的基因型。结果共繁殖出220只子代小鼠,经鉴定,NKCC1+/+小鼠占32%,NKCC+/-小鼠占50%,NKCC-/-小鼠占18%,其中,NKCC-/-小鼠生长较另外二型小鼠明显缓慢。结论采用Jack-on Laboratry提供的聚合酶链式反应法(PCR)可以成功地鉴别出不同基因表型的NKCC1基因敲除鼠,为研究NKCC1基因敲除小鼠相关课题提供了实验基础。
【Abstract】 Objective To breed and identify NKCC1 (Na~+-K~+-2Cl~- cotransporter) knock-out mice and to establish an animal experimental model for study.Methods NKCC1 knock-out mice were bred and reproduced under relative noise-free feeding conditions according to Mendel law. Genome DNA extracted from the mice tails was subjected to PCR for genotype identification.Results The experimental model for NKCC1 knock-out mice was successfully established and more NKCC1 knock-out mice were reproduced.Conclusion The polymerase chain reaction experiment method that Jackon Laboratry proposes can succeed in distinguishing different genetic NKCC1 gene of phenotype. Furthermore,it can lay out a solid foundation to further study NKCC1 knock-out mice correlation topic.
