PLGA与明胶膜支架材料对神经干细胞生长、分化影响的体外研究

【摘要】 目的:探讨PLGA作为中枢神经组织工程支架材料的可行性。方法:从胎龄14 ̄16d的Wistar大鼠分离培养获得神经干细胞,以108/L密度种植于PLGA膜、明胶膜和PLGA/明胶膜表面,置37℃、5%CO2培养箱中共培养,观察神经干细胞在PLGA膜、明胶膜和PLGA/明胶膜表面的生长、分化和PLGA的降解情况。结果:培养获得的神经干细胞表现具有自我更新和多相分化能力,能分化为神经元和胶质细胞。在观察期内PLGA膜无降解,神经干细胞在PLGA膜表面无贴附,但可在PLGA/明胶膜和明胶膜表面贴附、生长和分化。结论:PLGA可作为中枢神经组织工程支架材料。

【Abstract】 Objective: To explore the possibility of PLGA as a scaffold biomaterial for central nervous system regeneration. Methods: Neural stem cells were respectively seeded on the surfaces of PLGA, gelatin and PLGA/gelatin films at a density of 108cells/L. The growth and differentiation of neural stem cells and the degradation of PLGA were observed. Results: During the follow-up, PLGA didn’t degrade, no neural stem cells attached on the surface of PLGA films, but the cells attached, grew and differentiated on the surface of PLGA/gelatin and gelatin films. Conclusion: PLGA can be used for an ideal scaffold biomaterial for central nervous system regeneration.
