小鼠载脂蛋白CⅡ编码基因的克隆
Cloning DNA Encoding Sequence of Apolipoprotein CⅡin Mouse
【摘要】 目的:克隆小鼠载脂蛋白CⅡ(ApoCⅡ)编码序列。方法:采用逆转录-聚合酶链式反应(RT-PCR)的方法扩增获得ApoCⅡ编码序列,将ApoCⅡ基因编码片段重组入质粒pUC18,得到重组克隆载体pUC18-ApoCⅡ,进行序列测定。结果:经测序证实重组质粒pUC18-ApoCⅡ中的ApoCⅡ编码片段的序列与GenBank中的一致。结论:成功构建了含编码小鼠ApoCⅡ基因的重组克隆载体pUC18-ApoCⅡ,为基因工程生产有生物活性的ApoCⅡ、开展ApoCⅡ的免疫学测定及进一步探讨ApoCⅡ治疗高甘油三酯血症的可能性奠定了基础。
【Abstract】 Objective: To clone the DNA fragment encoding mouse apolipoprotein CⅡ(ApoCⅡ). Methods: The gene encoding ApoCⅡ was amplified by reverse transcription-polymerase chain reaction (RT-PCR). The amplified fragment was recombined into pUC18 plasmid and then sequenced. Results: The recombined plasmid pUC18-ApoCⅡwas obtained. The sequencing of ApoCⅡ in recombined plasma pUC18-ApoCⅡ was the same as GenBank sequence. Conclusion: The mouse ApoCⅡ was cloned successfully. The result provides new information for producing bioactive peptide of ApoCⅡ by genetic engineering, which may be useful in treating hypertriglyceridemia.
