大鼠神经干细胞和雪旺细胞体外共培养生长特性
Character of Co-culturing in Vitro of Rat’s Neural Stem Cell(NSC) and Schwann Cell (SC)
【摘要】 目的:探讨雪旺细胞(SC)对神经干细胞(NSC)的生物作用,为临床移植神经细胞提供依据和方法。方法:将神经干细胞分别在有雪旺细胞骨架、分泌物及活雪旺细胞饲养层的不同环境中培养,同时设立各自对照组,观察神经干细胞迁徙、贴壁能力、分化、细胞代谢、轴突生长、免疫组化神经丝染色情况。结果:与雪旺细胞有关的实验组神经干细胞大量分化为具有细长突起的细胞,神经丝(NF)标记阳性细胞多,突起长(特别是经胎牛血清培养过的雪旺细胞实验组),细胞贴壁力强,细胞生长代谢旺盛,突起粗壮并且生长速度快。结论:雪旺细胞能促进神经干细胞分化,并促进其生长。
【Abstract】 Objective: To investigate the biological effect of the Schwann cell to the neural stem cell and provide the basics and methods for clinical neural cell transplantation. Methods: The NSC was cultivated in SC skeleton, containing the secretion of SC and living SC respectively ,correspondingly the control groups were set up.The neural stem cell migration,differentiation,adhesion ,metabolism of cell , the development of axis and immunohistochemistry neurofilament dyeing were observed . Results: The groups of NSC containing Sc were greatly differentiated to the cells which had long neurite ,and neural fiber signed a lot of positive cells which had a long neurite(especially the SC cultivated by cow serum), high adhesion of cells ,vigorous metabolism ,more strong neurite and fast development. Conclusion: SC can facilitate the differentiation of NSC and prompt them to develop.
