肺气肿肺气虚证模型大鼠肺组织中基质金属蛋白酶表达的变化
CHANGES OF METALLOPROTEINASES EXPRESSION IN PULMONARY TISSUE OF RATS WITH EMPHYSEMA BELONGING TO DEFICIENCE OF LUNG QI SYNDROME
【摘要】 目的:观察基质金属蛋白酶-2(MMP-2)和MMP-9在肺气肿肺气虚证模型大鼠支气管和肺组织中的表达,探讨基质金属蛋白酶在肺气肿肺气虚证发病中的变化。方法:将SD大鼠90只随机分为对照1个月、2个月、3个月组和模型1个月、2个月、3个月组,采用气管内注入脂多糖(LPS)和烟熏双因素复合制作肺气肿肺气虚证大鼠模型,采用免疫组织化学染色SABC方法,检测肺组织中MMP-2和MMP-9的表达。结果:模型组大鼠支气管和肺组织中MMP-2和MMP-9在造模1个月后开始表达,并逐步升高,至造模3个月时表达明显增多,表达强度明显增强,各个月模型组与同时段对照组相比,均有显著差异。结论:MMP-2和MMP-9在肺气肿肺气虚证模型大鼠支气管和肺组织表达增多,活性增强。在肺气肿形成、发展、炎症持续方面起了重要作用,是肺气肿肺气虚证发病的主要机制之一。
【Abstract】 Objective: To observe the changes of metalloproteinases(MMP-2,MMP-9) expression in pulmonary tissue of emphysema with lung qi-deficiency syndrome ras, approach the significance and probable mechanism as well. Methods: 90 rats were divided into two groups: the control group(one month, two month and three month) and the emphysema with lung qi-deficiency syndrome group( one month, two month and three month). The rat’s model was built by intratracheal instillation of small dose lipopolysaccharide twice, exposure to cigarette smoking. The expression of MMP-2 and MMP-9 were examined by immunohistochemical method. Results: The expression of MMP-2 and MMP-9 were markedly increased in bronchi and pulmonary tissue in model group of each month compared with those of healthy control group. Conclusion: The increasment of MMP-2 and MMP-9 might play an important role in the development of the emphysema with lung qi-deficiency syndrome
