细胞色素P450 2J2基因启动子区单核苷酸多态性与高血压及脑卒中的相关性

【摘要】 目的研究中国汉族人细胞色素P450 2J2(CYP2J2)基因启动子区CYP2J2*7(G-50T)多态性分布频率及其与高血压和脑卒中的相关性。方法采用聚合酶链反应-限制性内切酶片段长度多态(PCR-RFLP)方法,对260例脑卒中患者、364例原发性高血压(EH)患者及年龄、性别与之相近的243例健康对照者进行CYP2J2*7多态性分析。结果脑卒中组、EH组及正常对照组CYP2J2*1(野生型)等位基因频率分别为0.963,0.962和0.957;CYP2J2*7等位基因频率分别为0.037,0.038和0.043。CYP2J2*7等位基因频率与高血压和脑卒中无明显相关性(P>0.05)。三组之间3种等位基因型分布频率无明显差异(均P>0.05),其中CYP2J2*1三组分别为92.69%、92.86%和91.36%;CYP2J2*7/*1分别为6.92%、6.87%和8.64%;CYP2J2*7分别为0.38%、0.27%和0。结论CYP2J2*7等位基因频率与高血压和脑卒中无明显相关性。

【Abstract】 Objective To evaluate the relationship between single nucleotide polymorphisms(SNPs) at position 50 in the promoter region of cytochromes P450 2J2(CYP2J2) gene and hypertension and stroke in Chinese Han population.Methods The fragment containing CYP2J2~*7(G-50T) was analyzed by using polymerase chain reaction-restriction fragment length polymorphisms(PCR-RFLP) in 260 cases of stroke and 364 cases of essential hypertension(EH) and 243 cases of health controls.Results The frequency of the CYP2J2~*1(wild type) in stroke,EH and control people was 0.963,0.962 and 0.957 respectively.The frequency of the CYP2J2~*7 in stroke,EH and control people was 0.037,0.038 and 0.043 respectively.The association between type or frequency of the G-50T polymorphism and EH or stroke was not found.Conclusion No association is inferred between the G-50T polymorphism and EH or stroke.
