孕期膳食锌缺乏下向调控胎盘IGF-Ⅱ和GLUT-1 mRNA的表达

【摘要】 目的探讨孕期膳食锌缺乏对胎盘IGF-Ⅱ和GLUT-1 mRNA表达的影响。方法36只SD孕鼠随机分为喂饲AIN93G配方饲料的正常对照组(ZC)、锌缺乏组(ZD)、配对喂养组(PF)以及国产商业饲料对照组(GC)。分别于孕9.5 d、孕13.5 d和孕17.5 d取母鼠血清、肝脏及胎盘,以半定量聚合酶链反应测定胎盘中IGF-Ⅱ和GLUT-1 mRNA以及肝脏MT-1 mRNA的相对表达量,以原子吸收法测定血清锌浓度。结果孕9.5 d和孕13.5 d时,ZD组血清锌浓度显著低于PF组、ZC组和GC组。孕17.5 d时,ZD组血清锌与PF组和ZC组相比无统计学差异。孕9.5 d和孕13.5 d时,ZD组母鼠肝脏MT-1 mRNA的相对表达量均显著低于PF组、ZC组和GC组。孕9.5 d、孕13.5 d和孕17.5 d时,ZD组胎盘IGF-Ⅱ和GLUT-1 mRNA相对表达量均显著低于ZC组、PF组和GC组。胎盘GLUT-1 mRNA的表达呈现出随着孕龄延长而增高的趋势。孕17.5 d时,PF组GLUT-1 mRNA的表达量与ZC组(P<0.05)和GC组(P<0.01)相比差异显著。结论孕期膳食锌缺乏显著下向调控胎盘IGF-Ⅱ和GLUT-1 mR- NA的表达。

【Abstract】 Objective To investigate the effect of dietary zinc deficiency on the expression of IGF-II and GLUT-1 mR-NA in placenta of rats during pregnancy. Methods 36 healthy pregnant SD rats were randomly divided into four groups: normal control (ZC), zinc deficient (ZD), pair-fed (PF) groups fed imported AIN93G formula diets and control group (GC) fed commercial diet made in China. Rats in all groups drink deionized distilled water. Placenta and maternal liver were taken at gestational 9.5 d, 13. 5 d and 17.5 d, respectively. The expression of IGF-II, GLUT-1 and MT-1 mRNA was examined by RT-PCR and the serum zinc concentrations ware measured by atomic absorption method. Results At gestational 9. 5 d and 13. 5 d, the zinc concentrations in group ZD were significantly lower than those in Groups PF, ZC and GC, respectively. The relative expression of MT-1 mRNA in the maternal livers from group ZD at gestational 9.5 d and 13.5 d was lower than those in groups PF, ZC and GC. At9.5d, 13.5d,17.5d, the expression of placental IGF-II and GLUT-1 mRNA in the group ZD was significantly lower than those in groups PF, ZC and GC, respectively. The expression of placental GLUT-1 mRNA increased with the pregnancy. At 17. 5 d, the expression of placental GLUT-1 mRNA in group PF was also significantly lower than those in groups ZC and GC. Conclusion Dietary zinc deficiency during pregnancy significantly down-regulated the expression of IGF-II and GLUT-1 mRNA in placenta of pregnant rats.
