高效液相色谱法分离邻、对位硝基甲苯同分异构体

【摘要】 利用高效液相色谱法对邻硝基甲苯和对硝基甲苯2种同分异构体的分离作了系统的研究.选择十八烷基键合相色谱柱COSMOSIL5C18-AR-Ⅱ(6.0mm×250mm),甲醇作为样品溶剂,甲醇和水的混合物作为流动相.确定了分离的最佳色谱条件为室温、V(甲醇)∶V(水)=70∶30、检测波长278nm、流动相流速1.0mL/min.回收率验证结果显示,准确度和灵敏度均符合分离要求.

【Abstract】 In this paper,the separation of p-nitrotoluene and o-nitrotoluene was studied comprehensively by high performance liquid chromatography.By comparison,C18 bonded-phase chromatography column of COSMO- SIL 5C18-AR-Ⅱ(6.0 mm×250 mm)was selected,methanol as solvent of specimen and mixture of methanol and water as mobile phase.The best chromatogryphy conditions are:room temperature,V(methanol):V(wa- ter)=70:30,detected wavelength is 278 nm and velocity of flow 1.0 mL/min.In addition,the method was validated by recoveries experiment.The results show this method is reproducible and sensitive enough for sepa- ration of p-nitrotoluene and o-nitrotoluene.
