毛细管电泳-化学发光法用于兔血中贝诺酯代谢物的分析

【摘要】 建立了一种用于药物在体内微量代谢物分析的毛细管电泳与化学发光在线联用方法,采用自制毛细管电泳-化学发光联用装置,对家兔血液中贝诺酯的活性代谢物扑热息痛进行了分析。考察了各种影响因素后,得到优化的电泳分离条件。在优化条件下,监测了贝诺酯灌胃后家兔血液中扑热息痛的含量变化。扑热息痛含量在贝诺酯灌胃后4 h达到峰值,扑热息痛的回收率为90%~115%,峰面积的RSD为2.1%~2.9%。本方法快速简单,检测灵敏度高,干扰小,适用于生物样品的分析。

【Abstract】 An analytical method based on on-line coupling of capillary electrophoresis with chemiluminescence detection was established to monitor the active metabolites of benorilate in rabbit plasma.The optimized condition in electrophoresis was determined after evaluating various controlling factors: Running buffer was 12 mmol·L-1 borax and 4 mmol·L-1 sodium hydroxide containing 5 mmol·L-1 β-cyclodextrin and 5 mmol·L-1 luminol,running voltage was 13 kV,the sample solution was injected 3 s with electrokinetic mode.Under the optimized conditions,the concentration of paracetamol in rabbit plasma in different time was determined.The linear range of paracetamol in rabbit plasma was about(0.1-)5.0 mg·L-1,with the determination limit of 3.81×10-2 mg·L-1(signalto-noise ratio is 3).The average recoveries and RSD(n=4) for peak area were 90%-115% and 2.1%-2.9%,respectively.The characteristics of this method are high speed,simply and sensitive,it is suitable for the analysis of biological samples,to determine the drug′s concentration in plasma.
