痹肿消汤对胶原诱导性关节炎大鼠滑膜白细胞介素10mRNA的影响

【摘要】 目的观察痹肿消汤对胶原诱导性关节炎大鼠白细胞介素10(IL-10)mRNA的影响,从基因水平探讨IL-10mRNA在类风湿关节炎的作用及痹肿消汤对类风湿关节炎抗炎作用。方法将34只大鼠随机分为3组,皮下注射II型胶原蛋白配合完全弗氏佐剂诱导类风湿关节炎模型,采用半定量RT-PCR法测定胶原诱导性关节炎大鼠滑膜细胞IL-10mRNA的表达。结果IL-10mRNA在正常组表达少量;25d模型组表达强于正常组(P<0.05),45d模型组表达弱于正常组(P<0.05),模型组25d表达高于45d;痹肿消汤组的IL-10mRNA表达强于模型组IL-10mRNA表达(P<0.05),在痹肿消汤组,45d表达稍高于25d,无统计学意义(P>0.05)。结论在胶原诱导性关节炎大鼠中痹肿消汤能增加IL-10mRNA的表达,提示痹肿消汤在治疗类风湿性关节炎中有免疫调节和抗炎作用,从而达到治疗作用。

【Abstract】 Objective To observe the effect of Bizhongxiao decoction on the expression of interleukin-10(IL-10) mRNA of C-II induced arthritis(CIA) in synovium of rat models for exploring pathogenesis in rheumatoid arthritis and antiinflammatory effect of BZXD on RA. Methods Thirty-four rats were randomly divided into 3 groups. The experimental rat models were established by subculaneous injection of type II collagen and complete Freund’s adjuvant. The expression of IL-10 mRNA in synovium of type II collagen induced arthritis was detected by the method of RT-PCR. Results IL-10mRNA expressed little or nothing in the normal group. In the model group, expression of IL-10mRNA on the 25~ th day was higher than that of the normal group(P<0.05), but expression of IL-10mRNA on the 45~ th day was less than that in the normal group(P<0.05). Expression of IL-10mRNA on the 25~ th day was higher than that of the 45~ th day in the model group (P<0.05). Expression of IL-10mRNA in the BZXD group was higher than that of the model group (P<0.05), while in the BZXD group, expression of IL-10mRNA on the 45~ th day was slightly higher than that of on the 25~ th day and showed no statistically significant difference (P>0.05). Conclusion BZXD can upregulate the expression of IL-10mRNA in CIA. It suggests that BZXD plays an important role in immunology regulation and antiinflammation effect in treatment of RA.
