固相萃取-反相高效液相色谱法测定火锅中吗啡和可待因
Determination of Morphine and Codeine in Hot Pot by Solid-phase Extraction and HPLC
【摘要】 目的采用固相萃取和反相高效液相色谱技术,建立简便、灵敏、准确的测定火锅汤料中吗啡和可待因含量的方法。方法样品经ChemElut固相萃取小柱纯化后,以甲醇/乙酸铵(pH7.0)(4060)溶液为流动相,在Shim-packCLC-ODS柱上进行洗脱分离,流速1.0ml/min。检测波长为212nm。结果吗啡浓度在0~50mg/L,可待因在0~80mg/L范围内具有良好的线性,检测下限分别为0.1mg/L和0.5mg/L。方法回收率分别为94.7%和92.3%,RSD分别为3.87%和4.58%。结论该方法灵敏度高、操作简便、快速,适用于实际样品的测定。
【Abstract】 s: Objective To establish a simple, sensitive and exact method for the determination of morphine and codeine in hot pot by solid-phase extraction and HPLC. Methods The hot pot samples were extracted and condensed by solid-phase extraction with Chem Elut catridges and then separated on a Shim-pack CLC-ODS HPLC column (6.0mm×150mm) using a gradient mobile phase consisting of CH3OH-20mmol/LCH3COONH4(pH7.O)/40:60 at a flow rate of 1.0 ml/min. Detection wavelength was 212nm. Results The liner range was 0~50mg/L and 0~80mg/L for morphine and codeine and the detection limits were 0.1mg/L and 0.5mg/L respectively. Extraction recoveries were 94.7% and 92.3%. The RSDs were 3.87% and 4.58%. Conclusion This method is sensitive, rapid and simple, and it can be used for the determination of morphine and codeine in hot pot samples.
