IFN-γ对TNF-α促OSF成纤维细胞增殖的抑制作用

【摘要】 目的观察γ-干扰素(interferon-γ,IFN-γ)对肿瘤坏死因子-α(tumornecrosisfacter-α,TNF-α)促正常人颊粘膜成纤维细胞(NM-FB)及口腔粘膜下纤维化(oralsubmucousfibrosis,OSF)患者颊粘膜成纤维细胞(OSF-FB)生长增殖的影响,探讨IFN-γ用于治疗OSF的可能性。方法取6例OSF患者及5例正常人颊粘膜,行成纤维细胞原代培养并传代,采用MTT比色法观察IFN-γ对TNF-α作用下NM-FB及OSF-FB的生长增殖的影响。结果IFN-γ对TNF-α促NM-FB及OSF-FB的生长增殖有明显抑制作用。结论IFN-γ是OSF形成过程中的一种负性调节因子,有可能用于临床治疗OSF。

【Abstract】 Objective To investigate the inhibition effects of IFN-γ on TNF-α enhanced proliferation of oral submucous fibrosis(OSF)-derived fibroblasts to see if IFN-γ can be used as a therapeutic regimen for OSF. Methods Primary fibroblast cultures were established from the buccal mucosa of five normal persons and six patients with OSF; and the effects of IFN-γ on the growth of TNF-α enhanced fibroblasts growth and proliferation by MTT colorimetric assay in vitro were studied. Result IFN-γ reduced the TNF-α enhanced growth and proliferation of oral mucosal-derived fibroblasts from both the OSF patients and the normal subjects. Conclusions The results show that IFN-γ is a cytokine which down-regulates effects on the TNF-α enhanced growth and proliferation of fibroblasts derived from patient with OSF, and suggest that IFN-γ might be play an important role in prevention and treatment of OSF.
