原花青素抑制佐剂性关节炎大鼠PGE2合成的机制
Inhibition mechanism of proanthocyanidins on the PGE2 product of rats suffered with adjuvant arthritis
【摘要】 目的研究原花青素对佐剂性关节炎大鼠体内PGE2合成的抑制机理。方法建立大鼠佐剂性关节炎模型,EIA法测大鼠足爪炎症组织中PGE2的含量,RT PCR和Western blot分别检测大鼠腹腔巨噬细胞内COX 2 mRNA和蛋白的表达。结果原花青素显著降低佐剂性关节炎大鼠足爪炎症组织中的PGE2含量,下调大鼠腹腔巨噬细胞COX 2 mRNA和蛋白的表达。结论原花青素对佐剂性关节炎大鼠体内PGE2合成的抑制作用可能与其抑制COX 2 mRNA和蛋白表达有关。
【Abstract】 Objective To study the inhibition mechanism of proanthocyanidins on the PGE2 product of rats with adjuvant arthritis(AA).Methods The adjuvant arthritis model of rats was induced by complete freunds adjuvant.The contents of PGE2 in paws of rats were measured by EIA.The expression of COX-2 mRNA and protein in rat′s peritoneal macrophages was analysed by RT-PCR and Western blot.Results Proanthocyanidins could significantly suppress the contents of PGE2 in edema paws and inhibit the expression of COX-2 mRNA and protein in peritoneal macrophages of rats suffered with adjuvant arthritis.Conclusions Inhibition mechanism of proanthocyanidins on the PGE2 product of rats with adjuvant arthritis may be related to inhibiting the expression of COX-2 mRNA and protein.
