复方降香胶囊的抗心肌缺血作用

【摘要】 目的观察复方降香胶囊(RCC)对犬实验性心肌梗死和大鼠急性心肌缺血的保护作用以及对小鼠常压耗氧量的影响。方法通过结扎犬左冠状动脉前降支产生急性心肌梗死模型,行心外膜电图标测和定量组织学N BT染色法,研究给药后对心肌缺血的影响;大鼠采用异丙肾上腺素(Iso)所致急性心肌缺血模型,观察给药后心电图的变化,测量给予Iso后心肌超氧化物歧化酶(SOD)活性与丙二醛(MDA)的含量;小鼠给予Iso后,观察常压整体耗氧量。结果RCC能显著降低冠脉结扎犬ST段升高总毫伏(mV)数(ΣST)和ST段升高总点数(N ST),减小N BT染色的梗塞面积,拮抗Iso引起的大鼠急性心肌缺血心电图ST段移位程度,增强缺血心肌SOD活性、降低MDA含量,降低小鼠常压下的耗氧量。结论RCC能对冠脉结扎犬的心肌梗死产生明显保护作用,具有抗Iso引起的大鼠急性心肌缺血作用和降低小鼠耗氧作用。

【Abstract】 Objective To observe the protective effects of capsules of rosewood compound(RCC)(traditional Chinese medicines) on acute myocardial ischemia in rats,myocardial infarction in experimental dogs,and oxygen consumption in normobaric hypoxia in mice.Methods Models of acute myocardial infarction were prepared by ligating the left anterior descending coronary artery of a dog.The extent of myocardial ischemia and infarction was determined by means of epicardial mapping and N-BT staining.The acute myocardial lesion caused by isoprenaline-induced ischemia was established and the protective effects were observed by electrocardiography.The superoxide dismutase(SOD) and malonic aldehyde(MDA) were measured after administration of isoprenaline,meanwhile,the oxygen consumption was observed in mice after intra venous administration of isoprenaline.Results It was shown that the increase of Σ-ST and N-ST,and the range of myocardial infarction caused by ligating coronary artery in dogs were obviously reduced by RCC.The changes of ST segment induced by isoprenaline in rats were also inhibited.The activity of SOD was increased,while the content of MDA was decreased.We also found that the oxygen consumption was reduced in mice.(Conclusions) RCC exerts protective effects on the myocardial infarction induced by ligating coronary artery in dogs as well as the acute myocardial ischemia induced by pituitrin in rats,furthermore,the oxygen consumption is reduced in mice by RCC.
