正交试验法优选急肝乐胶囊的提取工艺
Selection of optimum extraction process for Jiganle capsules by orthogonal design
【摘要】 目的优选急肝乐胶囊的提取工艺。方法川芎、公英先于80℃热水保温浸泡1 h,再与大黄、茵陈一起,利用正交试验法,考察100℃水浴煎煮时间、加水量、煎煮次数3因素、3水平对提取工艺的影响;以结合大黄素、结合大黄酚、阿魏酸为考察指标,用RP HPLC法测定各指标。结果结合大黄素、结合大黄酚总量的最佳提取工艺为100℃水浴煎煮15 min、加水量为药材重的12倍、煎煮2次,其中100℃水浴煎煮时间对结合大黄素、结合大黄酚的总量有非常显著性影响,加水量对其有显著性影响,煎煮次数影响不显著。阿魏酸的最佳提取工艺为100℃水浴煎煮20 min、加水量为药材重的12倍、煎煮3次,其中100℃水浴煎煮时间对阿魏酸的影响显著,加水量和煎煮次数没有显著性影响。结论急肝乐胶囊的最佳提取工艺为川芎、公英先于80℃热水保温浸泡1 h,再与大黄、茵陈一起,100℃水浴煎煮15 min,加水量为药材重的12倍,煎煮2次。
【Abstract】 Objective To find out the optimum extraction process for the preparation Jiganle capsules(traditional Chinese medicines).Methods Firstly,put Rhizoma Chuanxiong and Herba Taraxaci in water of 80 ℃ and soaked for one hour,then mixed with Radix et Rhizoma Rhei,Herba Artemisiae Capillaris.The three factor and three level orthogonal design was used.Boiling time,the amount of water and boiling times were defined as factors of the design,the yield of ferulic acid,combined chrysophanol and combined emodin was taken as the assessment index,HPLC was used to in the experiments.Results The more effective way to extract combined emodin and combined chrysophanol was boiling twice,15 minutes each time with 12 times amount of water.The results showed that boiling times had significant effect and the effect of boiling times was not significant.The more effective way to extract ferulic acid was boiling the mixture three times,20 minutes each time with 12 times amount of water.The results showed that boiling time affected prominently,boiling times and the amount of water affected not significantly.Conclusions The optimum extraction process for Jiganle capsule was that mixing Rhizoma Chuanxiong and Herba Taraxaci together in water of 80 ℃ for one hour,then boiling with Herba Artemisiae Capillaris and Radix et Rhizoma Rhei for twice,15 minutes each time with 12 times amount of water.
