【摘要】 目的研究抑癌基因RASSF1A在肝癌组织以及肝癌细胞株中的表达情况及其与临床相关因素间的关系。方法收集24例手术切除的肝癌及其癌旁组织以及4株肝癌细胞株(SMMC7721、HepG2、BEL7402和BEL7703(由国家人类基因组南方中心提供)。用RT-PCR和Northern Blot的方法来分析基因RASSF1A在肝癌组织和肝癌细胞株中的表达情况。结果67%的原发性肝癌组织中未检测到RASSF1A的mRNA,而相应的癌旁组织中仅33%的样本未表达RASSF1A;75%(3/4)肝癌细胞株也未表达RASSF1A。结论RASSF1A在肝癌中是一潜在的抑癌基因,RASSF1A基因的失表达在原发性肝癌的发生发展中起重要作用,检测RASSF1A的表达情况可用于对肝癌的早期发现和早期诊断,结合临床相关因素对肝癌预后评估也有帮助。更多还原

【Abstract】 Objective To study on the expression of tumor suppressor gene RASSF1A in hepatocellular carcinoma(HCC) and the correlation with clinical factors.Methods Twenty-four primary hepatocellular carcinoma and adjacent noncancerous tissue were obtained from 24 liver cancer patients by surgical resection.Four human HCC cell lines(SMMC7721,HepG2,BEL7402,and BEL7703) were provided by Chinese Human Genome Center(CHGC) in Shanghai.RT-PCR and Northern Blot were performed for quantitative analysis of the expression of tumor suppressor gene RASSF1A in HCC.Results RASSF1A mRNA was not detected in 67%(16/24) of the HCC,so was in 33%(8/24) of adjacent noncancerous tissues.Three of four(75%) HCC cell lines missed the expression of RASSF1A.Conclusion Testing for RASSF1A expression should become useful in HCC early detection and diagnosis and aid estimation of prognosis by combining the observation of potential clinical status of HCC progression.更多还原
