头皮药物注射疗法对大鼠缺氧缺血性脑损伤修复作用

【摘要】 目的研究头皮药物位点注射对缺氧缺血性脑损伤(HIBD)后脑细胞的修复作用。方法选取40日龄Wistar大鼠32只。其中5只为正常对照;27只制成HIBD动物模型,20 d后随机分为3组(n=9):A组用VitB1及VitB12额顶叶处注射,1次/d,共25 d;B组功能训练(Bobath法);C组不予处理。各组进行临床观察,并于干预结束7 d后处死动物,取脑组织进行光镜和电镜观察。结果1.临床观察:B与C组均有明显运动、记忆、行为异常和毛发改变;A组则无明显变化;2.光镜观察:B与C组均较A组神经细胞嗜酸性变严重,慢性变性神经细胞数量明显增多(P均<0.05);B与C组比较无明显差异(P>0.05)。3.电镜观察:B与C组缺血区暗细胞明显增多,细胞缩小并有空泡出现,A组改变较轻。结论HIBD模型可产生类似脑瘫的病理过程,药物注射疗法可明显改善神经细胞的损伤,产生修复作用。

【Abstract】 Objective To study the repairing effects of epicranium medication injection treatment on brain cells of hypoxic-ischemic brain damage(HIBD) and its mechanism.Methods Thirty-two Wistar rats of 40 day-old were selected,5 of which used as normal control group,27 used as test animal models of HIBD.Twenty days later,they were divided into 3 groups randomly(n=9):Group A were treated with VitB1 and VitB12 by injecting over frontal lobe and apical lobe once a day,for 25 days;Group B were treated with functional training(Bobath methods);Group C: no treatment.After 7 days treatment,all animals in three groups were killed and their brains were taken.The pathological changes of nerve cells,vascular endothelial cells,colloid cells and auxiliary fibers were observed under light microscope and electronic microscope.Results 1.Clinical observation: animals in group B and C all had obvious dyskinesia,hypomnesia,behavior disorder and pelage changed,and those in group A had no obvious change;2.Observed by light microscope: group B and C all had more serious nerve cell eosinophil denaturation,and the amount of chronic degenerating nerve-cell increased obviously than those in group A(P<0.05).There was no significant difference between group B and C(P>0.05);3.Observed by electronic microscope: hypoxic-ischemic area of group B and C were found,the size of cells became smaller and had vacuole appearance,and the stromatin of cells became condensed.But group A had less these changes.Conclusions HIBD model can form patho-process similar to cerebral palsy.Medication injection treatment relieve the injury of nerve cells and has better repairing effects.The experiment offers some theoretical evidence for epicranium medication injection treatment on cerebral palsy.
