宫内窒息对胎鼠脑组织型纤溶酶原激活物表达的影响
Effect of Intrauterine Asphyxia on Expression of Tissue-Type Plasminogen Activator in Fetal Rat′s Brain
【摘要】 目的观察宫内窒息后胎鼠脑内组织型纤溶酶原激活物(tPA)的表达情况。方法妊娠18 d孕鼠随机分为宫内窒息组和假手术组,通过结扎子宫动脉和胎盘分支血管15 min使胎鼠宫内窒息。分别在术后不同时间点取出胎鼠脑组织,进行tPA免疫组织化学染色。结果术后4、72 h宫内窒息组tPA阳性细胞数与假手术组相比均无统计学差异(P均>0.05),术后12、24、48 h组间比较均有统计学差异(P均<0.05);且术后24 h宫内窒息组阳性细胞表达达高峰。结论宫内窒息可诱导胎鼠脑内tPA表达增加。
【Abstract】 Objective To evaluate the expression of tissue-type plasminogen activator(tPA)in fetal rat′s brain after intrauterine asphyxia.Methods Pregnant rats on gestation 18 days were randomly divided into two groups: intrauterine asphyxia group and sham(operation) group.Intrauterine asphyxia was induced by ligating the uterus arterial and ramus vascular of placenta for 15 minutes.Then the fetal rat′s brains were taken out to be given immunohistochemistry of tPA.Results There were no significant difference of the number of tPA positive cell between the two groups at 4 hours and 72 hours after operation(all P>0.05),but there were significant difference between the two groups from 12 hours to 48 hours after operation(all P<0.05),and the expression of tPA got peak at 24 hours after(ope)-ration.Conclusion Intrauterine asphyxia can increase the expression of tPA in fetal rat′s brain.
