【摘要】 目的探讨肿瘤坏死因子-α(TNF-α)对肺泡Ⅱ型上皮细胞株A549增殖及凋亡的影响。方法将2003年2月至2004年1月中国医科大学附属第一医院放射肿瘤科实验室的肺泡Ⅱ型上皮细胞株A549用TNF-α处理后计算存活细胞数,四甲基偶氮唑盐(MTT)法测定细胞增殖能力,流式细胞仪检测细胞凋亡。结果TNF-α浓度和时间依赖性地抑制肺泡上皮细胞的增殖;TNF-α培养肺泡Ⅱ型上皮细胞12~48h后,凋亡的细胞数(%)明显高于对照组。结论TNF-α通过抑制肺泡Ⅱ型上皮细胞增殖和促进其凋亡,损伤肺泡上皮细胞,导致肺损伤。更多还原

【Abstract】 Objective To study the effect of TNF-α on the proliferation and apoptosis of type Ⅱlung alveolar epithelial cells line A549.Methods After the exposure of TNF-α to type Ⅱ lung alveolar epithelial cells line A549,the amounts of survival cells and apoptosis cells were observed.The proliferation of cells was measured by MTT and the apopotosis of cells was measured by the flow cytometric assay.Results TNF-α inhibited the proliferation of alveolar epithelial cells concentration and time reliably.The number of apoptosis cells(%)were much more than control group of 12~48 h after the exposure of TNF-α to type Ⅱlung alveolar epithelial cells.Conclusion TNF-α can damage alveolar epithelial cells and affect their function by inhibiting the proliferate of type Ⅱlung alveolar epithelial cells and increasing their apoptosis.更多还原
