【摘要】 目的探讨自体外周血干细胞移植及重组人粒细胞集落刺激因子(rhG-CSF)动员对糖尿病下肢动脉闭塞症患者血浆血管内皮生长因子(VEGF)及转化生长因子β1(TGF-β1)的影响。方法对2004-07~2005-06大庆油田总医院15例糖尿病下肢动脉闭塞症患者,应用rhG-CSF600μg每日皮下注射,连续5d,第5天采集外周血干细胞并制成悬液,将干细胞悬液肌肉注射入患者双下肢。于动员前后及移植后2个月,用ELISA方法检测血浆VEGF及TGF-β1质量浓度。结果与健康体检老人相比,患者动员前VEGF及TGF-β1质量浓度均显著增高(P=0·015及P=0·040)。患者移植后2个月血浆VEGF及TGF-β1,与动员前相比差异无显著性意义。结论糖尿病下肢动脉闭塞症患者血浆VEGF及TGF-β1质量浓度显著高于健康人。自体外周血干细胞移植及rhG-CSF动员不改变糖尿病下肢动脉闭塞症患者血浆VEGF及TGF-β1质量浓度,不增加血管增殖性病变的风险。更多还原

【Abstract】 Objective To investigate the change of plasma level of VEGF and TGF-β₁ in patients with diabetic lower limb arteriosclerosis obliterans by transplantation of autologous peripheral blood stem cells and rhG-CSF mobilization.Methods From July 2004 to June 2005,15 patients in Daging Oil-field General Hospifal were given rhG-CSF 600 μg/d for 5 days. Sooner the PBSC were intramuscularly injected into the two limbs of patients. Extracted peripheral blood 3 mL from patients before rhG-CSF mobilization and at the end of rhG-CSF mobilization and two months after autologous peripheral blood stem cells for examination of VEGF and TGF-β₁. Results The plasma level of VEGF and TGF-β₁,Compared with before rhG-CSF mobilization of patients groups,patients groups were significantly higher than normal oldsman groups.The plasma level of TGF-β₁ was（20.310±13.224）pg·mL -1 two months after autologous peripheral blood stem cells.Compared with before rhG-CSF mobilization it did not significantly change.Conclusion The plasma level of VEGF and TGF-β₁in patients with diabetic lower limb arteriosclerosis obliterans is significantly higher than of normal old.
