脱脂牛奶低温法保存微生物菌种的研究
Study of the laboratory strains frozen in degreased milk.
【摘要】 目的探索实验室常见菌种的最适合保藏方法。方法采用斜面低温法、半固体穿刺法、液体石蜡法、脱脂牛奶低温冻存法、冷冻干燥保存法进行研究,考察各方法保存后菌的存活时间和存活率。结果用斜面低温法、半固体穿刺法保藏6个月时菌种存活率在80%以上,但随后菌种迅速死亡存活率降低至0;液体石蜡法可保藏9~12个月。用脱脂牛奶低温冻存法保藏的菌种在2年后存活率仍达97.9%,且菌种的形态、生理特征基本未变。冷冻干燥保存2年后存活率100%。结论脱脂牛奶低温冻存法优于斜面低温法、半固体穿刺法等常规菌种保存法,保存2年后菌株存活率与冻干法比较无显著差异,且比冻干法操作简便,更容易开展。
【Abstract】 Objective To find the most convenient method for laboratory strains preservation.Methods To compare the livability of the laboratory strains frozen in degreased milk, with that of other methods including pure culture covered with olefin in 4 °C and lyophilized in degreased milk.Results 80% of the strains kept alive after 6 months with routine preserving methods, while 9~12 months kept in olefin. 97.9% of the strains frozen in degreased milk were alive after 2 years and lyophilized strains were 100%.Conclution Frozen in degreased milk was better than other routine preserving methods. It kept the same livability compared to lyophilized method, but more convenient.
