胱硫醚β合成酶基因在小鼠卵巢的定位表达分析
Localization of cystathionine-β-synthase gene in mouse ovary
【摘要】 目的了解胱硫醚β合成醚(cystathionine-β-synthase,CBS)在卵巢中的定位表达模式,并推测其在卵母细胞发育中生物学功能。方法利用mRNA原位杂交和 RT-PCR的方法检测CBS在成年小鼠卵巢中的表达。结果原位杂交结果显示,CBS在卵泡细胞中广泛表达;在初级卵泡细胞中,CBS阳性杂交信号强于次级卵泡以及窦卵泡的细胞; 在窦卵泡内部,CBS阳性信号既存在于近窦腔的颗粒细胞,也存在于卵丘细胞中。与经过孕母马血清(PMSG)作用的窦卵泡相比,未受PMSG作用的窦卵泡中,CBS阳性杂交信号尤其集中在靠近窦腔的颗粒细胞以及卵丘细胞。而在卵泡中的整个发育过程中的卵母细胞,都未发现有CBS的表达。利用RT-PCR的方法,以培养的各级卵泡进行的表达分析也证实了CBS表达的广泛性和卵母细胞中不表达的现象。结论 CBS参与了卵母细胞发育成熟的整个过程; 其在卵泡细胞中表达而在卵母细胞中不表达的特点说明了CBS是通过卵泡细胞与卵母细胞的相互作用而影响卵母细胞的生长发育的。
【Abstract】 Objective To understand the biological function of cystathionine-β-synthase gene (CBS) during oocyte development. Methods Detailed localization of CBS in ovary and oocyte was carried out by in situ hybridization and the expression profile of CBS in follicles was validated by reverse transcriptase PCR (RT-PCR). Results CBS was widely expressed in various developmental follicles. The hybridization signal was stronger in primary follicle than in secondary and antral follicles. Within the antral follicles, the expression of CBS was present in cells close to antrum and cumulus cells. The hybridization signals were generally stronger in follicle cells close to the antrum and cumulus cells in antral follicles from mice untreated with pregnant mare’s serum gonadotropin (PMSG). In additional, no CBS signal was detected in various developmental oocytes using mRNA in situ hybridization and RT-PCR. Conclusion Our results show that CBS is involved in the development of oocyte and follicle. The specific CBS expression profile in follicle and oocyte indicates that it is by the interaction between oocyte and follicular cells for CBS to act on oocyte development.
