食管鳞癌中COX-2和CD44v6的表达及其临床意义
The expression and significance of cyclooxygenase-2 and CD44v6 in esophageal squamous cell carcinoma
【摘要】 目的:探讨食管鳞癌中COX-2和CD44v6的表达情况及其相关性。方法:采用SP免疫组化方法检测15例正常食管粘膜(NEM)和62例食管鳞癌(ESCC)中COX-2和CD44v6的表达。结果:在15例NEM中,COX-2和CD44v6的表达率均为6.7%(1/15),而COX-2和CD44v6在ESCC中的检出率分别为51.6%(32/62)和50%(31/62),两者在NEM和ESCC中的表达存在显著性差异(P<0.05)。在不同病理分级的标本之间,II级和III级ESCC中,COX-2和CD44v6的表达率分别明显高于两者在I级ESCC中的表达率(P<0.05),而II级和III级ESCC中,COX-2和CD44v6的表达率则均无显著性差异(P>0.05);在I级和II级ESCC中,COX-2和CD44v6则均有明显相关性(P<0.05),在III级的ESCC中,COX-2和CD44v6的表达则无明显相关性(P>0.05)。在不同浸润程度的肿瘤中,COX-2和CD44v6的表达均有显著性差异(P<0.05),两者的表达与淋巴结转移均有明显相关性(P<0.05);COX-2和CD44v6的表达在不同性别、不同年龄的ESCC中均无显著性差异(P>0.05)。结论:COX-2和CD44v6均与食管鳞癌的发生、发展和转移密切相关,COX-2和CD44v6在食管鳞癌的浸润和转移过程中可能相互影响或共同作用导致肿瘤的浸润和转移,COX-2可能成为治疗食管鳞癌以及预防肿瘤转移的一个新靶点。
【Abstract】 Objective:To examine the expression of cyclooxygenase-2 (COX-2) and CD44v6 and to investigate the relationship between them in esophageal squamous cell carcinoma (ESCC). Methods: The expression of COX-2 and CD44v6 in 62 ESCC samples and 15 normal esophageal mucosa (NEM) samples from Shaanxi Province was examined by Streptavidin-Peroxidase (SP) method. Results: The positive rates of COX-2 and CD44v6 were 51.6(32/62)and 50(31/62)respectively in ESCC and 6.7 respectively in NEM. There exited statistical difference for COX-2 and CD44v6 between NEM and ESCC respectively (P<0.05). The expression rates of COX-2 and CD44v6 in grade II and grade III of ESCC were higher than those in grade I respectively (P<0.05), but no statistical difference between grade II and grade III of ESCC (P>0.05). The relationship between COX-2 and CD44v6 in ESCC was statistical significance (P=0.021). The expression rates of COX-2 and CD44v6 had no statistical different in gender and age respectively (P>0.05) but had statistical correlation with invasion (P<0.05) and lymph nodes metastasis (P<0.05) . In different histology grades of ESSC, the relationship between COX-2 and CD44v6 was statistical significant in grade I and II (P<0.05) but was not in grade III (P>0.05). Conclusion: COX-2 and CD44v6 were closely correlats in ESCC, in addition COX-2 and CD44v6 might be involved in metastasis of ESCC. COX-2 seems to be a new target of treatment and prevention of ESCC.
