人HSP70真核表达载体的构建及在大鼠胶质瘤细胞内的表达
Construction of HSP70 eukaryotic expression vector and its expression in rat glioma cells in vitro
【摘要】 目的:构建人热休克蛋白70(HSP70)的真核表达载体,并在大鼠胶质细胞瘤C6细胞中进行表达。方法:构建了HSP70与PCDNA3.1真核表达载体pCDNA3.1-HSP70,以脂质体法转染,同时利用免疫组织化学方法检测HSP70在大鼠胶质细胞瘤C6细胞中进行表达。结果:成功地构建了真核表达载体pCDNA3.1-HSP70,采用脂质体法转染大鼠胶质瘤C6细胞后,成功检测到HSP70在大鼠胶质细胞瘤C6细胞中表达。结论:成功地将外原性HSP70基因转染到大鼠胶质瘤C6细胞中并进行表达,为HSP70在肿瘤免疫治疗中的应用提供了实验材料。
【Abstract】 Objective: To construct the HSP 70 eukaryotic expression vector which is expressed in rat glioma C6 cell. Methods: The HSP 70 gene was amplified with method of PCR and cloned into the eukaryotic expression vector PCDNA 3.1 to construct the PCDNA 3.1 -HSP 70 plasmid. The plasmid was transfected into rat glioma C6 cells. Results: the HSP 70 eukaryotic expression vector PCDNA 3.1 -HSP 70 was constructed and transfected into C6 cell successfully. Conclusion: The HSP 70 gene was transfected and expressed into C6 cell successfully, which laid a foundation for the research of HSP 70 in the tumor immunotherapy.
