荞麦RAPD指纹图谱的建立及在品种鉴定中的应用
Generation of RAPD in Buckwheat and Its Application in Cultivars Identification
【摘要】 从9个荞麦品种的幼苗叶片提取DNA,使用CTAB和PVP并结合纯化过程中在高盐(N aC l)下沉淀DNA,可除去样品中影响DNA纯度的多酚类化合物和多糖.共使用13个随机引物,扩增的DNA片段大小为3.0K b~0.8 K b.从9个材料的13组RAPD指纹图谱上可以看出甜荞和苦荞的图谱差异很大(相同条带数占总条带数的比例为21%).在13个引物中有10个引物(占全部引物的77%)在甜荞的RAPD图谱上显示了品种间的DNA多态性,而苦荞只有6个引物(占全部引物的46%)产生了这种多态性.从荞麦RAPD指纹图谱上得到的特异带可以作为品种鉴定的分子标记.
【Abstract】 DNA was isolated from young leaves of nine buckwheat cultivars.The method used was a CTAB-based on extraction procedure including the use of NaCl to remove polyphenols during DNA purification.RAPD markers were generated by 13 arbitrary primers.The size of the amplified DNA fragments was from 800 to 3000 base pairs.The RAPD fingerprints of Fagopyrum esculentum were clearly different from that of F.tartaricum.They shared a small proportion(21 %) of RAPD markers from the 13 primers used in this survey,which implied a lower degree of homology among them.10 out of the 13 primers(77 %) revealed polymorphism in the F.esculentum cultivars,but only 6 primers(46 %) showed polymorphism within the F.tartaricum cultivars.The 9 cultivars could be separated by specific profiles resulting from 2~3 primer amplification.
