黄热疫苗病毒17D株在Vero细胞上的适应性传代
Adaptive Passage of Yellow Fever Vaccine Virus 17D Strain in Vero Cells
【摘要】 目的建立黄热疫苗病毒17D株的Vero细胞适应株。方法将2批鸡胚黄热疫苗病毒在Vero细胞上进行2次蚀斑筛选,并连续传代培养,然后对获得的4株Vero细胞适应株病毒YFV-Vero(5)-A1、YFV-Vero(5)-A2、YFV-Vero(5)-B1和YFV-Vero(5)-B2进行稳定性和免疫原性检测。结果4株Vero细胞适应株病毒可被黄热病毒(17D)猴免疫血清中和。在传代过程中,不同代次的病毒滴度稳定在6·90LgPFU/ml以上,高峰病变出现时间为5~7d。家兔和小鼠的免疫血清中和抗体效价分别为1∶640~1∶5120和1∶640~1∶2560。结论4株Vero细胞适应株病毒生物学性状稳定,免疫原性良好。
【Abstract】 Objective To obtain the adaptive strain of yellow fever vaccine virus in Vero cells.Methods Two lots of yellow fever vaccine virus were inoculated to Vero cells for plaque screening and further subculture.The obtained four adaptive strains,i.e. YFV-Vero(5)-A1,YFV-Vero(5)-A2,YFV-Vero(5)-B1 and YFV-Vero(5)-B2,were tested for stability and immunogenicity.Results The four adaptive strains were neutralized with monkey sera immunized with yellow fever virus 17D strain.During subculture,the titers of virus of various passages were more than 6.9 LgPFU/ml.The plaque sizes of virus of various passages showed no significant difference.The CPE of Vero cells reached a peak value 5-7 d after inoculation.The serum neutralizing antibody titers of rabbits and mice immunized with the adaptive strains were 1∶640-1∶5 120 and 1∶640-1∶2 560 respectively.Conclusion The four adaptive yellow fever virus 17D strains in Vero cells showed stable biological character and good immunogenicity.
