抗金黄色葡萄球菌及溶血性链球菌IgY的制备及临床疗效观察
Preparation and Clinical Effect of IgY against Aureus staphylococcus and Hemolytic Streptococcus
【摘要】 目的制备抗金黄色葡萄球菌及溶血性链球菌鸡卵黄IgG(IgY),并观察其治疗急性咽炎的临床疗效。方法将金黄色葡萄球菌及溶血性链球菌混合免疫母鸡,使用水稀释法、超滤法、西曲溴胺除脂法和硫酸铵盐析法提取、纯化IgY后,经全面检定,对110例急性咽炎进行临床疗效观察,治疗组60例,使用0·2%IgY口腔喷剂治疗7d;对照组50例,口服双黄连胶囊7d。结果IgY粗提物和纯化物纯度分别为41·6%和96·0%,效价分别达1∶1024和1∶8192;IgY粗提物在pH2·07~11·71条件下7d,效价基本不变;2·5~10·0mg/ml的纯化物抑制率为20·81%~68·11%。治疗组治愈率为26·67%,显效率为36·37%,有效率为30·00%,无效率为6·67%,总有效率为93·33%;对照组治愈率为12·00%,显效率为18·00%,有效率为44·00%,无效率为26·00%,总有效率为74·00%,差异有非常显著意义(P<0·005)。治疗组未见毒副作用。结论金黄色葡萄球菌及溶血性链球菌能够诱导母鸡产生特异性抗体,治疗急性咽炎疗效显著、安全。
【Abstract】 Objective To prepare the IgY against Aureus staphylococcus and hemolytic streptococcus and observe its curative effect on acute pharyngitis.Methods Immunize hens with the mixture of Aureus staphylococcus and hemolytic streptococcus,then extract and purify IgY from the eggs by dilution with water ultrafiltration,defating with cetrimide and salting out with ammonium sulfate.After overall control test,the IgY was observed for clinical effect in 110 cases of acute pharyngitis.The 60 patients in test group was treated with IgY stomat-spray,and the other 50 patients in control group with Shuang Huang Lian Jiao Nang(a capsule prepared with coptis rhizome),for 7 d.Results The purities of crude and purified IgY were 41.6% and 96.0%,and their titers were 1∶1 024 and 1∶8 192 respectively.The titer of crude IgY showed no significant change after storage at pH 2.07-11.71 for 7 d.The inhibiting rate of 2.5-10.0 mg/ml of purified IgY on the growth of Aureus staphylococcus was 20.81%-68.11%.The cure,significantly effective,effective,ineffective and total effective rates were 26.67%,36.37%,30.00%,6.67% and 93.33% in test group,and 12.00%,18.00%,44.00%,26.00% and 74.00% in control group,respectively(P<0.005).No adverse reaction was observed.Conclusion Both Aureus staphylococcus and hemolytic streptococcus induced IgY which was safe and effective in acute pharyngitis.
