番茄乙烯信号转录因子LeERF2多克隆抗体的制备

【摘要】 目的:作为番茄乙烯信号转录因子,LeERF2是在蛋白水平单独或同其他蛋白质协同与启动子或增强子相结合发挥其调控功能的,蛋白质的调控机制是信号转导领域中的关键研究内容。本研究目的是制备LeERF2抗体用于探讨番茄乙烯信号转录因子LeERF2在蛋白水平的生物活性及其生物学功能。方法:通过免疫新西兰大耳白雄兔和DEAE-52离子交换柱纯化分离制备了抗LeERF2多克隆抗体。结果与结论:抗体的效价大于10000,纯度达到了99%,Western印迹检测纯化后抗LeERF2多克隆抗体具有免疫特异性。

【Abstract】 Objective: Ethylene is an endogenous plant hormone that influences many aspects of plant growth and development. Control of ethylene responses is conducted by ethylene signal transduction pathway. Ethylene responsive element binding proteins(EREBPs) are transcriptional factors function in the ethylene signaling pathway in plants, helping to regulate ethylene responses by regulating genes transcription and expression. LeERF2 are identified as members of the EREBPs family, characterized by a conserved ERF domain. In order to further examine their regulation patterns, it is necessary to prepare anti-LeERF2 polyclonal antibody. Methods: Immunizing rabbits and purifying by DEAE-52 ion-column. Results & Conclusion: The titer of anti-LeERF2 serum was above 10 000, the purification level of polyclonal antibody was about 99% purified, anti-LeERF2 polyclonal antibodies had specific immunization function identified by Western blotting.
