蒲公英绿原酸提取分离工艺的研究
Study on Extraction and Separation Technology of Chlorogenic Acid in Dandelion
【摘要】 本文主要采用了正交分析的方法研究了从蒲公英中提取绿原酸,及采用大孔树脂NKA-9进一步分离绿原酸的最适条件,并采用制备性液相色谱的方法将其纯化。确定从蒲公英中提取绿原酸的最佳工艺参数为:物料比1:16,乙醇浓度80%,温度60℃,反应时间2h,绿原酸提取率可达60%以上。选用NKA-9树脂来分离绿原酸,确定其最适的分离条件是:上样浓度为0.26mg/ml,pH2.0,进样液体积/大孔树脂质量比值为12,流速为2ml/min,洗脱剂30%乙醇,绿原酸提取物纯度可达25.3%。最后采用半制备性液相色谱分离得绿原酸提取物,其纯度达到87.6%。
【Abstract】 Conditions of extraction and further isolation of chlorogenic acid from dandelion (Taraxacum. mongolicum Hand) in orthogonal test with macroporous resin NKA-9 were studied in this paper. Chlorogenic acid was isolated from dandelion with semi-preparing liquid chromatography. The optimum extraction conditions for dandelion chlorogenic acid were established as follows: ratio of material to liquor 1:16, 80% ethanol(V/V) as solvent and reflux extraction at 60℃ for 2 hours. The extraction rate of chlorogenic acid is as high as 69%. Chlorogenic acid was purified with macroporous resin NKA-9. The optimum purified conditions were established as follows: the sample concerntration 0.26mg/ml (pH 2.0), the ratio of the sample volume to the mass of macroporous resin NKA-9 12, and 30% ethanol as eluting solvent at 2ml/min. the purity of obtained chlorogenic acid is 25.3%. Finally, chlorogenic acid of higher purification (87.6%) is obtained with semi-preparing liquid chromatography.
