米曲霉β-半乳糖苷酶催化合成低聚半乳糖的工艺研究
【摘要】 研究了反应时间、乳糖浓度、温度、加酶量、pH对低聚半乳糖合成的影响,结果表明,加酶量、乳糖浓度、反应温度的影响较大。通过正交实验确定的米曲霉β-半乳糖苷酶催化合成低聚半乳糖的最佳工艺条件为:pH6.0、温度55℃、乳糖浓度40%、加酶量40U/g、反应时间32h。在最佳工艺条件下,低聚半乳糖的合成率为32.4%。
【Abstract】 The effects of time, initial lactose concentration, temperatur e, enzyme amount and pH on the synthesis of galacto-oligosaccharide from lactos e using β-galactosidase were investigated. The result showed as follow: initia l lactose concentration, enzyme amount, and temperature had strong impacts on the reaction and the galacto-oligosaccharide yield reached 32.4% under the optim um conditions: time was 32h, temperature was 55°C, the initial lactose concen tration was40%, pH was 6.0, and the enzyme amount was 40U/g.
