杂色曲霉素灌胃后小鼠脉络丛细胞超微结构和TNF-α表达的改变

【摘要】 本研究观察了杂色曲霉素(sterigmatocystin,ST)对小鼠脉络丛细胞超微结构和TNF-α表达的影响。给BALB/c小鼠一次灌胃ST3000μg/kg,分别于灌胃后0.5,1,2,4,8和16h处死动物,在扫描电镜下观察脉络丛细胞超微结构的改变,并且用原位杂交方法观察了脉络丛细胞TNF-α的表达。结果显示:ST灌胃1h后脉络丛细胞上出现火山口样改变,并且分泌颗粒增多,分泌颗粒在处理4h后最多,处理8h后分泌颗粒明显皱缩,处理16h后分泌颗粒明显减少。TNF-α在对照组和处理组各时间点脉络丛细胞均有表达,但处理组在ST灌胃0.5h后TNF-α表达迅速增加,4h达最高峰,之后开始下降,16h后下降到最低点。处理组与对照组比较,TNF-α表达显著增加(P<0.01)。上述结果表明,单次ST灌胃后对脉络丛细胞有一定程度的损伤,而TNF-α在脉络丛细胞损伤过程中可能起着重要作用。

【Abstract】 To investigate the effects of sterigmatocystin (ST) on the ultrastructure and the expression of TNF-α in choroid plexus cells after single administration of ST in BALB/c mice in vivo. The mice were treated intragastrically by gavage with ST at 3000 μg/kg. The ultrastructural changes and the expression of TNF-α at mRNA level in choroid plexus cells were studied with scanning electron microscopy (SEM) and in situ hybridization at 0.5, 1, 2, 4, 8 and 16 h after oral administration, respectively. The results showed that after oral administration of ST, secretary granules could be seen and volcanic crater like changes appeared in choroid plexus cells after ST treatment 1 h. The number of secretary granules peaked at 4 h, shrinked at 8 h and obviously decreased at 16 h. TNF-α mRNA expression could be found in some choroid plexus cells at every time point in control and treated groups. The number of TNF-α positive cells increased after ST treatment 0.5 h, reached its peak after ST treatment 4 h and then began to decrease. The positive rate and intensity of TNF-α mRNA expression were all significantly higher than those in control group (P<0.01). Our study revealed that oral ST exposure in mice could cause damaging changes of choroid plexus cells, and the TNF-α might play a critical role during the injury of choroid plexus cells.
