核团微量注射下丘脑视前区“针刺灌流液”对去卵巢大鼠整体效应的研究

【摘要】 目的本文采用针刺一组特定穴位后的去卵巢大鼠视前区(POA)推挽灌流液,微量注射于另一个去卵巢大鼠的下丘脑POA,观察其是否可作为一种针刺信息分子的载体,产生相应的(针刺)效应。为建立一种能在离体组织和培养细胞上,研究针刺治病信号传(转)导机制的可行的实验方法。方法采用切除双侧卵巢导致下丘脑-垂体-卵巢轴(HPOA)功能异常的大鼠模型。将动物分成收集灌流液组和注射灌流液组两大组。收集灌流液组又分为收集电针后下丘脑POA推挽灌流液(简称“针刺灌流液”)组(OVXAP)和去卵巢大鼠POA灌流液组(OVXP);OVXAP给予电针“关元”为主的一组穴位处理,以阴道脱落细胞学检查和R IA测定针刺后动物血雌二醇(E2),作为判断针效的指标;用HPLC测定“针刺灌流液”中的单胺类和氨基酸递质及β-内啡肽的含量作为质控标准,对“针刺灌流液”进行质量控制;注射灌流液组又分为注射“针刺灌流液”组(IOVXAP)、注射灌流液对照组(IOVXP)和注射人工脑脊液对照组(IaCSF)。注射前后分别作动物阴道脱落细胞学检查和血E2水平测定,以观察其是否可作为一种针刺信息分子载体,将针刺效应转移到新的个体,产生针刺样效应。结果IOVXAP大鼠血E2显著高于IOVXP和IaCSF(P<0.05),且IOVXAP大鼠阴道涂片出现大量成熟脱落细胞,而IOVXP和IaCSF未出现类似的变化。结论“针刺灌流液”可能作为一种针刺信息分子的载体,把针刺信息转移到其他个体上,模拟特定的针刺效应,为进一步研究针刺治病信号的传(转)导机制提供新的实验方法。

【Abstract】 Objective For study of the mechanism of the acupuncture signal conduction or transformation in the central never system,a new experimental method was established.Methods 30 SD female rats were ovariectomized as the animal models with dysfunction of hypothalamic-pituitary-ovarian axis(HPOA) and divided randomly into two major groups,collecting the perfusate group and microinjecting perfusate group.Collecting perfusate group were subdivided into collecting OVX+ EA perfusate group(OVXAP) and OVX perfusate group(OVXP).OVXAP was performed a push-pull perfusion in hypothalamic preoptic area(POA) with acupuncture treatment at Guanyuan(CV4),Zhongji(CV3) acupoints etc.for three days.An exfoliative cytology examination in vaginal smears was taken 6 hours later after last EA treatment,which was considered as an index of effective EA and the levels of blood estrogen(E2),the contents of GnRH and β endorphin in the acupuncture perfusate were determined by RIA and the levels of monoamine and amino acid neurotransmitters in the perfusate were detected by HPLC,which were adopted as the quality control standards of the acupuncture perfusate.OVXP was performed a push-pull perfusion in hypothalamic POA without EA treatment.Microinjecting perfusate group were subdivided into microinjecting acupuncture perfusate group(IOVXAP),microinjecting OVX perfusate group(IOVXP) and microinjecting artificial cerebrospinal fluid group(IaCSF).Before and after injection,the vaginal exfoliative cytology of each rat was examined for determination whether the acupuncture-like effect appeared.Results The blood E2 level in IOVXAP increased significantly compared with IOVX and IaCSF(P<0.05),a few of mature vaginal epithelia reappeared in IOVXAP,and the percent of mature exfoliative epithelia in vaginal smears were significant difference compared with IOVX and IaCSF(P<0.05).Conclusion Acupuncture perfusate might be play a role of acupuncture signal vehicle and conduct the acupuncture signal into the another animals in vitro to imitate the specific acupuncture-like effect,which might provide a new method for study of the mechanism of acupuncture signal conduction or transformation.
