益气活血方中黄芪不同剂量对缺血性脑损伤大鼠抗氧化作用的影响

【摘要】 目的:观察益气活血剂中不同剂量黄芪组方对缺血性脑损伤大鼠抗氧化作用的影响,以探讨黄芪适宜的使用剂量。方法:采用颈内动脉注射血栓的方法,复制多发梗死性脑缺血大鼠模型,观察益气活血剂中不同剂量黄芪(30g、60g、120g)组方对模型动物血浆SOD活性、MDA含量和脑组织SOD活性及MDA、NO含量的影响。结果:60g黄芪配伍组能明显升高SOD活性,降低MDA、NO含量,与空白对照组相仿(P>0.05),与模型组比较有显著性意义(P<0.05~0.01)。120g黄芪配伍组也能使SOD活性升高,MDA、NO含量降低,但与空白组比较,血浆SOD、MDA含量及脑组织NO的含量均有显著性差异(P<0.05~0.01);与模型组比较,脑组织NO含量、SOD活性均无显著性差异(P>0.05)。30g黄芪配伍组呈现出增加SOD活性、降低MDA、NO含量的趋势,但与空白对照组比较有显著性差异(P<0.05~0.01),与模型组比较无显著性差异(P>0.05)。结论:在治疗缺血性脑血管疾病的益气活血复方配伍中,黄芪的用量宜大,但随着其剂量的增大,其抗氧化效应反而减弱。其用量在人以60g/d左右为宜。

【Abstract】 Objective: To observe the effect of different dosages of Radix Astragali in prescriptions for invigorating Qi and activating blood circulation on antioxidation of rats with ischemic cerebral injury so as to determine the proper dosage of Radix Astragali. Methods: Internal carotid artery injection of thrombus is adopted to produce cerebral ischemia rat model with multiple infarction so as to observe the effects of different dosages of Radix Asteris (30 g, 60 g, 120 g) on plasma SOD activity, MDA content, brain tissue SOD activity, and the content of MDA and NO. Results: In the 60 gram group, SOD increased markedly, and MDA, NO content decreased, similar to the control group(P > 0.05), but with marked differences compared with the model group (P < 0.05~0.01). In the 120 gram group,SOD also increased, and MDA, NO content decreased, but compared with the control group, plasma SOD and MDA content and brain tissue NO content showed great differences (P < 0.05~0.01); compared with the model group, brain tissue NO content and SOD activity showed no great difference (P > 0.05). In the 30 gram group, there was a tendency for the SOD activity to increase, MDA and NO content to decrease, with marked differences from the blank control group (P < 0.05~0.01), but with no marked difference from the model group (P > 0.05). Conclusions: In the prescriptions for invigorating qi and activating blood on ischemic cerebrovasculra diseases, large dosage of Radix Asteris is recommended, but with the increase of the dosage, antioxidation effect is decreased. The appropriate dosage for humans is 60 g/d.
