益母草生物碱和黄酮成分抗大鼠心肌缺血药效学研究

【摘要】 目的:探讨益母草生物碱和黄酮成分抗大鼠心肌缺血的药效学作用。方法:将实验大鼠随机分为正常组、模型组、益母草生物碱组、黄酮组、丹参组,及生物碱加黄酮大、中、小剂量组。采用大剂量异丙肾上腺素(ISO)皮下注射,造成大鼠急性心肌缺血损伤模型,用药后检测各组大鼠Ⅱ导联心电图J点变化情况、心肌组织形态学变化、血流动力学变化及酶谱变化。结果:与生物碱组、黄酮组及小剂量组相比,益母草生物碱加黄酮大、大中剂量组J点未发生明显偏移(P<0.05),左室内压峰值(LVSP)、左室内压最大变化速率(±dp/dtmax)向正常水平恢复(P<0.05),抑制损伤大鼠血清中肌酸磷酸激酶(CPK)、乳酸脱氢酶活性(LDH)活性及降低缺血心肌组织中丙二醛(MDA)的含量,提高大鼠心肌组织中超氧化物歧化酶(SOD)的活性(P<0.05),组织切片进一步说明生物碱和黄酮成分能有效保护心肌组织(P<0.05)。结论:益母草中的生物碱、黄酮成分能有效抑制异丙肾上腺素ISO所致心肌组织缺血损伤。

【Abstract】 Objective: To observe the active ingredients of leonurus in myocardial ischemia induced by isoproterenol (ISO). Methods: The model of myocardial ischemia was induced by subcutaneous injection of ISO (4 mg/kg for two days, once a day).The experimental rats were randomly divided into normal group, ISO group, positive control group and leonurus composition groups. J Point in ECG, the histological changes of myocardium, the activity of LDH, CPK in serum and the level of MDA, SOD in myocardial tissue, and the LVSP, ±dp/dtmax were surveiled. Results: Compared with the model group, the leonurus composition groups had recovery effects on J point (ECG), blood pressure, LVSP, and ±dp/dtmax , with decreased level of LDH, CPK, MDA, and increased activity of SOD. The histopathological changes confirmed the protection effects of alkaloid and chromocor on the myocardial injury. Conclusion: The alkaloid and chromocor are the effective ingredients of leonurus heteropyllus sweet, which can alleviate the acute myocardial ischemia caused by ISO in rats.
