高通量检测技术检测乙型肝炎病毒变异的临床意义
Detection of hepatitis B virus gene mutations by high-throughput method and its clinical significance
【摘要】 目的探讨基因芯片技术检测乙型肝炎病毒(HBV)6位点变异的临床意义。方法对91例乙型肝炎患者采用基因芯片技术,检测HBV 6位点的自然变异。结果91例乙型肝炎患者HBV变异率98.9%(90/91),其中BCP区1762位点变异率最高(61.5%)。重度肝炎和肝硬化中双位点变异显著高于轻中度肝炎(P<0.01)。29例进行过拉米呋啶抗病毒治疗的病例中P区552位点变异率为96.6%(28/29)。结论基因芯片技术检测HBV变异具有高通量、高灵敏等特点,对观察HBV基因突变有很高的临床应用价值。
【Abstract】 Objective To study the clinical significance of the detection of hepatitis B virus(HBV) gene mutations by gene chip technique. Methods 6 sites mutations of HBV in 91 patients were detected by gene chip technique. Results The mutations of HBV existed in all the 91 patients except one(90/91),the rate of mutation was 98.9%.The mutation rate of BCP section 1762 site was the highest(61.5%).Double sites mutations in the patients of severe hepatitis and cirrhosis were significanly higher than in mild hepatitis(P<0.01).The mutation rates of P section 552 site were 96.6%(28/29) in 29 patients treated by lamivudine. Conclusions The gene chip technique is simple to perform and can be used as a rapid and high-throughput method.It has a high clinical value in detecting the HBV gene mutation.
