环氧合酶-2在幽门螺杆菌影响酸分泌机制中的作用
Effects of cyclooxygenase-2 in the mechanism of gastric acid secretion on Helicobacter pylori-induced gastritis in mice
【摘要】 目的通过幽门螺杆菌(Hp)感染的动物模型研究胃黏膜环氧合酶-2(COX-2)蛋白表达在酸分泌机制中的作用。方法清洁级BALB/c小鼠40只,随机分4组:①阴性对照组予0.4 mL 0.9%氯化钠溶液灌喂;②急性感染组予0.4 mL Hp菌液灌喂;两组均为5 d,最后1次灌喂2周后处死。③清除组和慢性感染组:予0.4 mL,Hp菌液灌喂,共5 d,最后1次灌喂2个月后分为清除组(予克拉霉素3.5 mg·kg-1·d-1,分2次灌喂,连续7 d)和慢性感染组(予0.9%氯化钠溶液,方法同清除组);1个月后分别处死两组动物。均取胃黏膜标本,采用免疫组织化学半定量法测定胃窦黏膜COX-2蛋白和H+-K+ ATP酶mRNA基因表达的变化。结果阴性对照组COX-2蛋白表达率为(3.25±1.50)%,显著低于急性感染组(11.38±2.93)%、慢性感染组(31.18±6.78)%和清除组(23.80±5.92)%(P值均<0.05)。4组的COX-2蛋白与H+-K+ ATP酶mRNA基因表达均不相关(P值均> 0.05)。结论Hp感染后COX-2蛋白表达增多伴H+-K+ ATP酶mRNA基因表达下降,清除Hp后COX-2蛋白表达下降伴H+-K+ ATP酶mRNA基因表达上升,但两者不相关。
【Abstract】 Objective To investigate the effects of cyclooxygenase(COX)-2 in the mechanism of gastric acid secretion on Helicobacter pylori (H. pylori)-induced gastritis in mice. Methods Forty 7-weeks old SPF BALB/C mice were divided into four groups. The mice in the acute infection group were inoculated orally H. pylori liquid (Sydney Strain 1, SS1) 0, 4 mL(1×109CFC) per mouse every day for five days while those of the H. pylori negative group were given placebo of equivalent quantity. Two weeks after the last inoculum, all mice were sacrificed. Anther 20 mice were inoculated orally H, pylori liquid per mouse every day for five days. Two months after infection of H, pylori, all mice were divided into two groups, the chronic infection group and the eradication group with each of 10 mice. The mice in the eradication group were administered clarithromycin individually (13. 5 mg·kg-1·d-1) twice per day for one week(one mouse died). Meanwhile, the mice in the infection group were given the same amount of placebo. All mice were killed one mouth after the administration. The gastric mucosae were removed for inmunohistochemical study of COX-2 protein and RT-PCR test of H+ -K+ ATPase mRNA gene expression. Results The relationship of COX-2 protein expression and H+ -K+ ATPase gene expression was analysed in the acute infection group and the control group, showingγ0. 121 vs 0. 312, P 0. 74 vs 0. 308, respectively. Studies in the chronic infection group and the eradication group, resultingγ0. 121 vs 0. 205, P 0. 74 vs 0. 598, respectively. The results showed no significant relations between COX-2 protein expression and H+ -K+ ATPase gene expression. Conclusion Under infection of H. pylori, COX-2 protein expression increases with association of decreasing H+-K+ ATPase mRNA gene expression. After eradication of H. pylori, COX-2 protein expression decreases but accompanied by increase of H+-K+ ATPase mRNA gene expression. Correlative analysis shows no relationship between them.
