【摘要】 目的从牛血浆中制备高活力及高收率的凝血酶。方法以牛血为原料,采用柠檬酸钡吸附、硫酸铵分级沉淀、DEAE-纤维素色谱等方法制备凝血酶原,并模拟生理条件用凝血酶原活酶复合物,包括牛凝血因子Ⅴ、Ⅹa、磷脂和Ca2+对凝血酶原进行激活。结果采用此方法制备的凝血酶的比活力为85 U/mg,收率为135μg/mL血浆。结论采用此制备技术,可以大规模制备高活力及高收率的牛凝血酶。更多还原

【Abstract】 Purpose To prepare high activity and high yield thrombin from bovine blood.MethodsUsing techniques of barium citrate absorption,ammonium sulfate fractionation and DEAE-cellulose chromatography to prepare bovine prothrombin.Prothrombin was activated by prothrombinase complex simulating the natural condition.The prothrombinase complex included coagulation factor Ⅴ,coagulation factor ,phospholipids and Ca²⁺.ResultsThe activity and yield were 85 U/mg,135 μg/mL plasma respectively.ConclusionUsing the above techniques can prepare high activity and high yield bovine thrombin on a large scale.更多还原
