底丘脑核深部脑电刺激对大鼠呼吸的影响
Effect of Subthalamic Nucleus-Deep Brain Stimulation on Breath Frequency of Rats
【摘要】 目的研究底丘脑核(STN)的深部脑电刺激(DBS)对大鼠呼吸的影响。方法24只SD大鼠随机分为4组(n=6):对照组(无刺激)、高频强电流组(130 Hz/100μA)、高频弱电流组(130 Hz/3μA)和低频强电流组(40 Hz/100μA)。利用大鼠脑立体定向仪将电极植入右侧STN,记录刺激前、刺激第5和第25分钟的呼吸频率。结果高频强电流组和高频弱电流组刺激前呼吸频率分别为(68.0±7.9)和(71.8±6.7)次/m in;刺激后第5分钟的呼吸频率分别为(57.8±10.6)和(65.8±7.8)次/m in;第25分钟的呼吸频率分别为(49.8±8.9)和(56.0±9.2)次/m in;同组各时间点的呼吸频率变化有统计学意义(P<0.05)。低频强电流组和对照组各时间点的呼吸频率无明显变化(P>0.05)。结论大鼠在高频率DBS下呼吸频率减慢,但与刺激强度关系不大。
【Abstract】 Objective To study the effect of subthalamic nucleus(STN)-deep brain stimulation(DBS) on breath frequency of rats. Methods Twenty-four SD rats were divided into four groups randomly: non-stimulation(control)group,high-frequency and high-intensity(130 Hz/100 μA)group,high-frequency and low-intensity(130 Hz/3 μA)group and low-frequency and high-intensity(40 Hz/100 μA)group.The target of DBS was STN which was located by stereotactic apparatus.Breath frequencies of rats were recorded at the time of pre-stimulation,5 and 25 min after execution of stimulation. Results In the 130 Hz/100 μA group,the breath frequency of rats was(68.0±7.9)/min at pre-stimulation,and decreased to(57.8±10.6)/min after execution of stimulation for 5 min,and to(49.8±(8.9))/min after execution of stimulation for 25 min.In the 130 Hz/3 μA group,the breath frequencies of rats at different stages were(71.8±6.7),(65.8±7.8) and(56.0±9.2)/min,respectively.There were significant diffe-(rences) on breath frequency among the different time points in the 130 Hz/100 μA group and the 130 Hz/3 μA group(P<0.05).However,in the control group and the 40 Hz/100 μA group,there were no significant differences on breath frequency at any stage of stimulation(P>0.05). Conclusion The effects of DBS on breath frequency of rats may be related to frequency rather than intensity of stimulation.
