Ⅳ型胶原糖基化对表皮干细胞生物学特性的影响

【摘要】 目的研究糖尿病患者皮肤组织中Ⅳ型胶原的糖基化对表皮干细胞生物学特性的影响。方法体外培养表皮干细胞,用免疫细胞化学和免疫荧光化学方法鉴定其标志物(角蛋白19和β1整合素),将表皮干细胞分别接种在包被有正常Ⅳ型胶原和糖基化Ⅳ型胶原的培养板上,比较两者黏附率、生长曲线和克隆生成率。结果糖基化Ⅳ型胶原上生长的表皮干细胞的黏附率和克隆生成率均较正常胶原组低(P<0.05)。结论Ⅳ型胶原的糖基化抑制了表皮干细胞的黏附与增殖,可能是糖尿病患者创面难愈的机制之一。

【Abstract】 Objective To investigate the effect of glycation of collagen Ⅳ on biological character of epidermal stem cells. Methods After epidermal stem cells were cultured,keratinocyte 19(K19) and integrin β1 were identified by immunocytochemistry and immunofluorescence.Then they were seeded on the culture plates which were coated with normal collagen Ⅳ and glycated collagen Ⅳ.The growth curve,cell adhesion rate and clone production rate were determined. Results The cell adhesion rate and clone production rate of glycated group were lower than those of normal group(P<0.05). Conclusion Adhesion and proliferation of epidermal stem cells were inhibited by glycation of collagen Ⅳ,which may be one cause for difficult healing of diabetic wound.
