内皮祖细胞定向分化平滑肌细胞的实验研究

【摘要】 目的内皮祖细胞是一种理论上非常合适的血管组织工程种子细胞,最终分化成成熟内皮细胞。实验拟证实内皮祖细胞可以定向分化成平滑肌细胞。方法以猪为实验动物,采集骨髓单核细胞,分离培养内皮祖细胞,然后应用血小板衍生生长因子(PDGF)BB诱导内皮祖细胞定向分化。通过免疫组化和免疫荧光法鉴定细胞类型。结果经过诱导的内皮祖细胞定向分化成平滑肌细胞。未经诱导的细胞分化成内皮细胞。结论内皮祖细胞不仅可以分化成内皮细胞,而且可以在PDGF-BB作用下分化成平滑肌细胞,是一种理想的血管组织工程种子细胞来源。

【Abstract】 Objective Endothelial progenitor cell is a potential seeding cell for vascular engineering. They would eventually differentiate into endothelial cells. The study was designed to identify that they could also trans-differen- tiate into smooth muscle cells. Methods Total mononuclear cells were isolated from bone marrow of pigs. Endothelial progenitor cells were cultured in endothelial growth medium, and then platelet-derived growth factor BB (PDGF-BB) was mixed into the medium and cultured for another 4 days and 10 days. Smooth muscle cell markers such as α-smooth muscle actin, calponin and main heavy chain were identified by imunocytochemistry and immunofluorescence. Results Uninduced cells were differentiated into endothelial cells and induced cells were partly differentiated into smooth muscle cells. Conclusion Endothelial progenitor cells can differentiate into endothelial cells and can be induced into smooth muscle cells by PDGF-BB. It could be used as an auto-donated seeding cell resource in vessel engineering.
