创伤后关节软骨中MMP-3的表达及意义
Expression of Matrix Metalloproteinase-3 in Rabbit Articular Cartilage after Trauma
【摘要】 目的探讨基质金属蛋白酶-3(MMP-3)在创伤后关节软骨退变中的作用。方法手术建立单纯软骨缺损的动物模型,术后4周、8周对软骨退变进行大体评分,用免疫组织化学方法检测MMP-3在关节软骨中的表达。结果术后关节软骨发生退变,MMP-3在关节软骨中的表达与退变有关。结论MMP-3在创伤后软骨退变过程中发挥重要作用。
【Abstract】 Objective To discuss the impacts of MMP-3 (matrix metalloproteinases) on degeneration of articular cartilage after trauma. Methods To make animal model: One partial-thickness articular cartilage defect of 4.0 mm in diameter was created in the trochlear articular cartilage of femoral condyles of right knee joint, and the opposite as pseudo operative controls. The severity of cartilage degeneration were observed under disseeting microscope 4 and 8 weeks respectively after operation Immunohistochemical and morphometric analyses were adopted to record the in situ expression and distribution of MMP-3 in cartilage at protein level. Results Cartilage degeneration around the defect 4 weeks after operation was signifieantly severer than that in control group (P<0.01). The degeneration after eight weeks was more significantly(P<0.01). The expression of MMP-3 increased significantly over the degeneration of articular cartilage in both femoral condyles and patella, and which was higher in patellar cartilage than in femoral condyles. Conclusion Cartilage degeneration will develop secondarily after trauma. MMP-3 is closely involoved in cartilage injury and degeneration.
