【摘要】 目的以套式RT-PCR方法检测癌胚抗原(CEA)mRNA在乳腺癌患者外周血中的表达,并探讨其在乳腺癌诊断与治疗中的意义。方法收集乳腺癌患者52例(乳腺癌组)、乳腺增生性病变患者40例(增生组)、健康查体者20例(对照组),采集外周血样本。分别用套式RT-PCR法检测其外周血中CEA mRNA的表达。结果乳腺癌组外周血CEA mRNA阳性率为40.4%,增生组及对照组分别为5.0%和0,两两比较均有显著差异(P均<0.01);乳腺癌组TNMⅢ期、Ⅳ期患者CEA mRNA阳性表达率明显高于Ⅰ期、Ⅱ期患者,外周血CEA mRNA阳性表达与淋巴结转移密切相关。结论外周血CEA mRNA表达可作为乳腺癌转移的一个辅助监测指标。更多还原

【Abstract】 [Objective] The present study is to investigate the carcinoembryonic antigen(CEA) mRNA expression of carcinoembryonic antigen(CEA) in peripheral blood from patients with breast cancer or breast hyperplasia,and to evaluate its clinical significance.[Methods] Peripheral blood samples were collected from 52 patients with breast cancer,40 patients with breast hyperplasia,and 20 healthy volunteers.CEA mRNA was detected by nested reverse transcription polymerase chain reaction(RT-PCR).[Results] The products of RT-PCR were analyzed by agrose gel electrophoresis.And their molecular weight was standardized by markers in the position of about 132 bp.The positive rate of CEA mRNA expression in blood samples of 52 patients with breast cancer was 40.4%.And no healthy volunteers was positive.But 2 of the 40(5.0%) patients with breast hyperplasia(5.0%) were positive.There were significant differences among three groups.The positive rate increasesd with the pathological staging and lymph node metastasis,but there were no significant relations between the rate and the malignant extent of cell differentiation.[Conclusion] The expression of CEA mRNA in peripheral blood cells correlates with the pathological staging and lymph node metastasis of breast cancer,and it can be of potential use to monitor the micrometastases of tumor.更多还原
