离体肾脏保存过程中bcl-2蛋白、bcl-2mRNA表达变化及FK506对其的影响
The change of bcl-2、bcl-2 mRNA expression in isolated renal preservation and the effect of FK506
【摘要】 目的探讨离体肾脏保存过程中bcl-2蛋白、bcl-2 mRNA表达及他可莫司(FK 506)对其的影响。方法将40只W istar雄性大鼠随机分为实验组和对照组各20只,建立离体肾脏模型。对照组将离体肾脏置于常规4℃UW保存液中;实验组在上液中加入FK 506,分别检测肾脏保存2、4、8、16 h后bcl-2蛋白、bcl-2 mRNA表达。结果实验组4、8、16 h后bcl-2蛋白、bcl-2 mRNA表达明显高于对照组(P<0.05)。结论FK 506可能通过上调bcl-2蛋白、bcl-2 mRNA表达,减轻离体肾脏保存过程中的损伤。
【Abstract】 [Objective] To investigate bcl-2,bcl-2-mRNA expression in renal preservation and effect of FK506.[Methods] 40 male Wistar rats were randomly divided into test group and control group,the model of the preservation of isolated rat renal was established.The isolated rat renal was put into 4℃ UW liquid in the control group and mixed FK506 in the test group.Renal tissue bcl-2、,bcl-2 mRNA expression were studied respectively with immunohistochemical staining,reverse transcription polymerase chain reaction(PT-PCR) at different preservation time points(2,4,8,16 h).[Result] The bcl-2,bcl-2 mRNA expression in test group were obviously increased(P<0.05).[Conclusion] FK506 may decrease the isolated rat renal injury through increasing bcl-2,bcl-2 mRNA expression.
