白藜芦醇苷对慢性低氧性肺动脉高压大鼠肺内突变型p53基因表达的影响

【摘要】 目的研究白藜芦醇苷(PD)对慢性低氧性肺动脉高压(CHPH)大鼠肺内突变型p53基因表达的影响。方法将SD大鼠随机分为对照组、低氧组和低氧+PD治疗组。观察PD对CHPH大鼠平均肺动脉压(mPAP)、右心室肥厚指数[右室/左室+室间隔(R/L+S)]、肺血管形态学指标、突变型p53基因表达的影响。结果21d后,低氧组mPAP、R/(L+S)、肺血管管壁面积占血管总面积的百分比、管壁厚度占血管外径的百分比及突变型p53基因表达明显升高,肺血管管腔面积占血管总面积的百分比明显降低;低氧+PD组上述变化得到抑制或减轻。结论PD可有效降低肺动脉压,延缓大鼠CHPH进程,其机理可能与抑制突变型p53基因表达及细胞增殖,从而有效抑制肺血管重建有关。

【Abstract】 Objective: To investigate the effects of polidatin on expression of p53 in chronic hypoxic rats. Methods: 30 healthy Spraque-Dawley male rats were randomly divided into normal control group, chronic hypoxic group and hypoxia plus polidatin treatment group. The model of rat chronic hypoxic pulmonary hypertension (CHPH) was made by intermittent isobaric hypoxia for 21 days. The indexs including mean pulmonary arterial pressure (mPAP), R/(L+S), vascular morphologic indexes of pulmonary and expression of p53 in pulmonary tissue of rats were measured. Results: 21 days after exposed to hypoxia,the mPAP, R/(L+S), MT%, MA%, expression of p53 increased significantly. VA% decreased significantly. Pretreated with PD could attenuate the changes mentioned above. Conclusions: Pretreatment with PD can effectively prevent the formation of CHPH. It may be related to the inhibition of p53 which can enhance proliferation and decrease apoptosis, that contribute to postpone the development of pulmonary vascular remodeling.
