腰椎间盘细胞培养方法探讨
【摘要】 目的寻找使椎间盘细胞更具生物学活性的培养方法。方法将25例胎儿及成人腰椎间盘细胞分为A组(足月胎儿)、B组(18~24岁)、C组(25~34岁)、D组(35~44岁)、E组(45~55岁),均分别进行单层培养和立体培养,培养检测Ⅰ、Ⅱ型胶原(CⅠ、CⅡ)的含量。结果C、D、E组立体培养较单层培养C、C含量明显增高(P均<0.05)。结论立体培养细胞具有更大的生物学活性,有助于椎间盘细胞生物学的进一步研究。
【Abstract】 Objective: To detect the content of collagen type Ⅰ and type Ⅱ of intervertebral disc cells cultured by mono-layer and three-dimension methods respectively, and find the more better method of disc culture. Methods: 25 cases of foetuses and adults disc cells were divided into A, B, C, D and E group and cultured with nono-layer and three-dimension methods, the content of collagen type Ⅰ and type Ⅱ of disc cells were detected. Results: Among the cells cultrured with three-dimension method, the level of collagen type Ⅰ and type Ⅱ was higher than those among monolayer cultue cells in group C, D and E (P<0.05). Conclusion: Three-dimension cultured cells show more higher biological activity than monolayer cultured cells, the former would be more helpful to the study of intervertebral disc cell biology.
