rhBMP-2/卵磷脂复合物治疗股骨头坏死的实验研究
Experiment study of the composite of rhBMP-2 and lecithin on avascular necrosis of femoral head
【摘要】 目的观察人重组骨形态发生蛋白-2(rhBMP-2)/卵磷脂复合材料修复早期兔股骨头坏死(ANFH)病灶清除后缺损区的成骨能力,为临床治疗股骨头坏死提供新方法。方法采用液氮冷冻制备兔双侧股骨头坏死动物模型,实验侧植入rhBMP-2/卵磷脂复合材料,对照侧植入单纯卵磷脂片;3、6、9周时取股骨头标本进行组织学检查及碱性磷酸酶(ALP)活性、Ca2+测定,评价rhBMP-2/卵磷脂复合材料成骨效果。结果3周时实验侧缺损区可见成骨细胞和骨细胞,9周有大量的骨细胞和成骨细胞,对照侧仅见纤维组织充填;ALP活性及Ca2+含量实验侧均高于对照侧。结论rhBMP-2/卵磷脂复合材料具有良好的诱导成骨活性,生物相容性好,可降解,能够有效的提高ANFH的骨修复能力。
【Abstract】 Objective: To observe the effect of the composite of recombinant human bone morphogenetic protein 2(rhBMP-2)and lecithin in repairing bone defects of avascular necrosis of the femoral head in rabbits to examine the activity of rhBMP-2, and to explore a new method in treatment of avascular necrosis of the femoral head. Method: The models of bilateral avascular necrosis of the femoral head were established in rabbits by using freezing method with liquid nitrogon. Composite of rhBMP-2 and lecithin was infilled into the experimental side and lecithin was infilled into the control side. The osteogenic function was evaluated by histology,alkaline phosphate and calcium. Results: The new bone formed and elongate into the infilling areas on the 3th week after operation,on the 9th week the new bone trabecular was distributed in all over the infilling areas in experimental side. There was more new bone formation in the experimental side than that in the control side. The activity of alkaline phosphate and level of calcium in the experimental side were much higher than those of control side. Conclusion: Composite of rhBMP-2 and lecithin has a strong activity of osteoinduction and fine biocompatibility and biodegradability, can improve the repairing ability effectively of ANFH.
