【摘要】 目的探讨自大鼠外周血中获取内皮前细胞(EPC)的方法。方法SD大鼠皮下注射粒细胞集落刺激因子(G-CSF)后抽取外周动脉血,密度梯度离心法获取单个核细胞,用完全培养液在体外培养后进行CD31、CD34、FLK-1和vW F免疫荧光染色,并检测其结合UEA-1、摄取acLDL的能力。结果将此方法收获的细胞进行CD 31、CD 34、FLK-1和vW F免疫荧光染色并检测其结合UEA-1、摄取ac-LDL的能力,证实为EPC。结论G-CSF刺激、密度梯度离心、完全培养液法可自大鼠外周血中获取较多EPC。更多还原

【Abstract】 Objective:To explore the procedures of isolation,cultivation and purification of endothelial progenitor cells(EPC)from peripheral bloods.Method:Peripheral blood was obtained from the femoral artery of SD rat.Mononuclear cells were isolated by Ficoll density gradient centrifugation,cultured in a special medium and stained immunofluorescence.Result:Cells obtained by this procedure were immunohistologically proved EPC.Conclusion:Relatively purified EPC can be obtained by certain procedure of isolation and culture.更多还原
