巨细胞病毒感染人脐静脉内皮细胞后RAGEmRNA的时序性表达及其意义

【摘要】 目的:研究人巨细胞病毒(HCMV)感染对人脐静脉内皮细胞(HUVEC)糖基化终产物受体(RAGE)表达的影响,探讨HCMV感染致动脉粥样硬化的作用机制。方法:用HCMV感染HUVEC,采用RT-PCR方法检测不同感染时段细胞RAGEmRNA的表达。结果:HCMV感染HUVEC后,0 h时RAGEmR-NA有基础水平的表达,4 h开始升高,8 h时表达水平明显升高,随感染时间延长,表达量逐渐增高,感染24 h时达高峰,48 h开始回落,72 h表达量仍维持在较高的水平,显著高于0 h。各时段与0 h比较,均有统计学意义(P<0.01)。结论:HCMV感染人脐静脉内皮细胞后能够促进RAGE mRNA的表达,并且呈时间依赖性。HCMV有可能通过上调RAGE介导内皮细胞的炎症反应,促进动脉粥样硬化的发生、发展。

【Abstract】 Objective: To study the mechanims of cytomegalovirus infection by investigating the sequential changes of the receptor for advanced glycation end products in human umbilical vein endothelial cells with human cytomegalovirus infection.Methods: HUVEC was cultured and then infected by HCMV.Expression of RAGE mRNA was detected by reverse transcriptase polymerase chain reaction.Results: RAGE mRNA was expressed at a low level in control group,and began to increase 4 hours after HCMV infection,rose to a significant level at 12 hours,reached the peak at 24 hours.After 48 hours,it declined obviously and remained a relatively high level at 72 hours(P<0.01).Conclusions: HCMV infection can enhance the expression of RAGE mRNA in HUVEC in a time-effect fashion,and may induce the inflammatory reaction by up-regulating RAGE expression on endothelial cells,which facilitate the occurrence and development of atherosclerosis.
