霉酚酸酯对高糖所致大鼠肾小球系膜细胞增殖的影响

【摘要】 目的:观察霉酚酸酯(mycophenolate mofetil MMF)对高糖环境大鼠肾小球系膜细胞(mesangialcells MCs)增殖及Ⅳ型胶原的影响。方法:以30 mmol/L葡萄糖刺激体外培养的大鼠MCs增殖,加入不同浓度的MMF,采用MTT比色法观察6、12、24、48 h系膜细胞的增殖情况;放射免疫法测定细胞上清液中Ⅳ型胶原的含量。结果:葡萄糖对系膜细胞的增殖效应呈浓度和时间依赖性,反应最佳浓度为30 mmol/L,最佳时点为24 h。MMF有抑制高糖所致MCs增殖、减少细胞外基质的作用,并呈剂量时间依赖性。结论:高糖在一定浓度和一定时段内有刺激大鼠系膜细胞增殖的效应,MMF对高糖所致MCs增殖具有抑制作用,且随MMF剂量增大,作用时间的延长,抑制作用增强。

【Abstract】 Objective:To investigate the effect of mycophenolate mofetil(MMF)on high glucose-induced proliferation and the type Ⅳ collagen in rat mesangial cells(MCs).Methods:Rat MCs were stimulated by different concentrations of high glucose,out of which 30?mmol/L was fit for the stimulation concentration,and then different concentrations of MMF were added into MCs of glucose induced rat.The MCs proliferative rate within 6,12,24 and 48 hours was detected by MTT colorimetry.The type Ⅳ collagen in supernatant contents of medium was detected by radioimmunoassay method.Results: The MCs proliferation presented time-and dosedependence by different concentrations of high glucose,which got to the best at the concentration of 30?mmol/L and at 24 hours.MMF could inhibit the high glucose-induced proliferation and showed time-and dose-dependence.Conclusion: High concentration of glucose can stimulate MCs and results in MCs proliferation by time-and dose-dependence,meanwhile MMF can inhibit such a high glucose-induced proliferation of MCs,also showing time-and dose-dependence.
